HAIR CYCLE

(A) Graphic stages of hair follicle (HF) morphogenesis: hair placode (initiation of HFs pattern formation), immature hair germ, hair peg, and fully developed and differentiated anagen hair by the end of first postnatal resting phase of telogen. Mature HF is built of a reservoir of hair follicle Stem Cells (hfSCs) which is connected above to an associated sebaceous gland (SG), located below the responsive hair germ (HG) with underlying dermal papilla (DP). During the cyclic hair growth, each HF undergoes phases of telogen (quiescence, resting), anagen (activation, regrowth), and catagen(degeneration).  (B) During the growth phase, the hair bulb cells comprise actively proliferative matrix cells which undergo final differentiation creating an inner root sheath (IRS) and hair shaft (HS, protruding out of the skin surface, visible part of HF) layers of the HF mini-organ. HS, hair shaft is composed of Ch, the cuticle of the hair shaft; Co, cortex of hair shaft; Me, medulla; DP, dermal papilla; IRS, inner root sheath contains; He, Henle’s layer; Hu, Huxley’s layer; Ci, cuticle; Cp, companion layer; ORS, outer root sheath.

Wnt/β-catenin signaling pathway

Wnt/β-catenin signaling mechanism. 
(A) In unstimulated cells, β-catenin forms a macromolecular complex with Axin, APC, CK1α, and GSK3 (destruction complex). Phosphorylation by CK1α and GSK3 prepares β-catenin to be ubiquitinated and degraded in the proteasome 26S. 
(B) The interaction of Wnt glycoprotein ligands with Frizzled receptors and LRP5/6 co-receptors recruits DVL to the plasma membrane. Then, GSK3 phosphorylates LRP5/6 and β-catenin is no longer phosphorylated and ubiquitinated because the destruction complex disassembles. The β-catenin accumulated in the cytoplasm is translocated to the nucleus where it displaces the co-repressor Groucho and interacts with TCF to activate gene expression.

Caffeine:(Topical : 1%, Mesotherapy : up to 0.1%)
Caffeine is an alkaloid methylxanthine and functions as a phosphodiesterase inhibitor, promoting cellular proliferation.  Caffeine counteracts the inhibitory effects of testosterone on hair growth, promotes hair shaft elongation, prolongs anagen duration, and stimulates hair matrix keratinocyte proliferation . Caffeine also downregulates testosterone-induced transforming growth factor (TGF)-β1 expression, a hair growth inhibitor, and increases expression of insulin-like growth factor (IGF)-1, a hair growth promoter. Female hair follicles appear to have a higher sensitivity to caffeine.

Aminoacids: 
N-acetyl-L-cysteine (NAC) : Cysteine plays a central role in hair health; it forms dimers that are oxidized to produce cystine, creating disulfide bridges that provide strength and rigidity between keratin strands.
Lysine is thought to play a role in the absorption of iron.
Methionine is vital for both keratin and procollagen synthesis.
Arginine is protective against the negative effects of hydrogen peroxide on hair proteins and hair surface lipids secondary to oxidative coloring or bleaching.

Vitamins & Minerals

Vitamin A: This vitamin helps to produce a substance called sebum. Sebum is created by the scalp, and it acts as a natural conditioner. It keeps hair from breaking off or drying out and becoming itchy.
Dexpanthenol(Vit B5): is involved in collagen synthesis, one of the major components of hair, reduces sebum secretion, and helps create an environment where hair can grow well. It also strengthens weak hair follicles, helps retain moisture, and prevents hair loss caused by dry scalp.
Biotin(Vit B7): Biotin is a B-complex vitamin and cofactor for carboxylase enzymes involved in fatty acid synthesis, amino acid catabolism, gluconeogenesis, and mitochondrial function in hair root cells
Folate(Vit B9): Folate (or folic acid, the synthetic version of folate) helps renew cells that promote hair growth.
Vitamin C: Vitamin C is an antioxidant, and it helps the absorption of iron into the body.
Vitamin D: Vitamin D receptors are intracellular receptors expressed in hair follicles, essential for normal hair cycle and differentiation of the interfollicular epidermis.
